According to the Zhitong Finance App, Insili Intelligence (03696) announced that the first human phase I clinical trial data of ISM6331, an innovative pan-TEAD inhibitor driven by artificial intelligence, was received at the 2026 European Society of Medical Oncology (ESMO) annual meeting and made a brief oral presentation (Rapid Oral Presentation).

This phase I clinical trial was conducted simultaneously in China and the United States to evaluate the safety, tolerability, pharmacokinetics, pharmacodynamics, and initial antitumor activity of ISM6331 in patients with advanced or metastatic malignant mesothelioma or other solid tumors.

ISM6331 is a potent pan-TEAD inhibitor that not only effectively targets tumors with abnormal Hippo pathways, but also shows synergistic anti-tumor effects and overcomes drug resistance in combination therapy. ISM6331's innovative non-covalent structure is guided by Chemistry42's structure-based drug design strategy.

In preclinical studies, ISM6331 showed a wide range of anti-tumor efficacy, strong activity at low doses, ideal safety, and good ADMET characteristics. In August 2024, ISM6331 was granted Orphan Drug Qualification (ODD) by the US Food and Drug Administration for the treatment of mesothelioma; its global multi-center phase I clinical trial completed its first patient administration in January 2025.
